Dose-response curve in antipsychotic-exposed GaMg cells extrapolated to therapeutically relevant serum levels. HMGCR expression levels changes for each drug, following transformation of the molar concentrations used in the cell culture experiment into multiples of their therapeutically relevant serum levels (see below). Due to effects on cell viability, the highest concentrations of chlorpromazine, haloperidol and ziprasidone in the culturing media was 25 μM. Typical and atypical antipsychotic drugs are denoted with open and filled symbols, respectively. Note the logarithmic scale on the x-axis. The upper therapeutic serum concentration for each drug was set according to the AGNP-TDM expert group consensus guidelines [36]: clozapine 2000 nM, chlorpromazine 1000 nM, olanzapine 250 nM, haloperidol 50 nM, risperidone 140 nM and ziprasidone 250 nM.
