Schematic illustrating biopanning protocol to generate generic TDP-43 detection phages. To isolate detection phages that bind all healthy and disease related TDP-43 variants, we first eliminated all phage particles that bind non-desired targets including BSA and aggregated alpha-synuclein using immunotubes. Next, in the positive biopanning phase, the remaining phage particles were added to a mica surface containing TDP-43 immunoprecipitated from the motor cortex of healthy brain tissue. Any bound phages were glycine eluted and added to a mica surface with TDP-43 immunoprecipitated from ALS brain tissue. Following glycine elution the same manner of positive panning was repeated for TDP-43 immunoprecipitated from FTD brain tissue. Eluted phages were sequenced to verify scFv integrity and larger batches of each produced for further analysis
