Actin dynamics mediate leptin and insulin activation of K ATP A, representative cell-attached recording from an acutely isolated ARC neurone with leptin (10 nM) in the electrode solution. Following attainment of increased KATP activity, bath application of jasplakinolide (100 nM) reversibly reduced channel activity. The corresponding diary plot for this experiment is shown. B, representative cell-attached recording from an ARC neurone. Bath application of insulin (10 nM) increased KATP channel activity and subsequent bath addition of 100 nM jasplakinolide, concomitant with 10 nM insulin, reversibly inhibited the insulin stimulated KATP activity.
