TUNEL Staining. To further explore the difference in the cell death response that was observed in the LDPT neurons compared to what was found in our previous TPS study, we utilized a TUNEL assay kit to examine for signs of cell death 1-week following T9 level contusion. A, C, E , DTMR retrogradely labelled TPS neurons probed with the TUNEL assay kit show colocalization of a TUNEL labelled nucleus within a retrogradely labelled TPS neuron, additionally, multiple TUNEL labelled nuclei are present in the merged image, possibly glial cells or interneurons, not retrogradely labelled by TMR. B, D, F , However, when cervical spinal cord is examined, while DTMR retrogradely labelled LDPT neurons are clearly visible, no signs of TUNEL labelling are observed in neurons, or in the surrounding tissue.
