Effect of over-expression of GFP-PRDX6 on ROS levels in RGCs following hypoxic stress. Cellular extract was prepared and resolved on SDS-PAGE, and Western analysis was performed using antibody specific to PRDX6, to assess both exogenous and endogenous expression of PRDX6. ~55 kDa protein band was detected as recombinant GFP-PRDX6 protein (A, right lane). Histogram showing ROS levels in RGCs after exposure to hypoxia (B&C). Cells over-expressed with GFP-PRDX6 prevented excessive generation of ROS against CoCl2 (50, 100 or 200 μM). Cells were transiently transfected with either GFP vector or GFP-PRDX6. After 24 h cells treated with CoCl2 (50, 100 or 200 μM).
